How do the shrines work? Sometimes They don’t give any points at all after activating them.

You have to do something around them until a bell/chime rings and it starts giving you a buff with a visual pulsing aura.
I think it’s kill a certain amount of the oncoming waves while in its vicinity. All I know for sure is just activating it doesn’t do it.
i noticed that i couldn’t finish my prayer as long as there were even minor enemies around, even as ogryn. i held the use button but the progress bar would disappear about midway. when the shrine had been cleansed of enemies, one can finish the prayer, a gong is heard, and the shrine glows golden. that should be a clear sign.
and btw, it’s easier to wait at the shrine, kite the boss + captain who spawns around it, and lay toxin there. not run ahead, into other enemies, and have those and the bosses to fight.
